Description

Rat monoclonal [M/K-2] to VCAM1

Specificity

Present on the +/+ 2.4 stromal cell clone, large cells in bone marrow, and a subset of thymic macrophages. The stromal cell antigen is also detectable on the EOMA and BMS2 endothelial cell lines. Reacts with all known isoforms of murine VCAM-1. Murine vascular adhesion molecule (VCAM1), single chain glycoprotein of Mr100-kDa (under reducing conditions).

Target

Function

Important in cell-cell recognition. Appears to function in leukocyte-endothelial cell adhesion. Interacts with the beta-1 integrin VLA4 on leukocytes, and mediates both adhesion and signal transduction. The VCAM1/VLA4 interaction may play a pathophysiologic role both in immune responses and in leukocyte emigration to sites of inflammation.

Tissue specificity

Expressed on inflammed vascular endothelium, as well as on macrophage-like and dendritic cell types in both normal and inflammed tissue.

Sequence similarities

Contains 7 Ig-like C2-type (immunoglobulin-like) domains.

Domain

Either the first or the fourth Ig-like C2-type domain is required for VLA4-dependent cell adhesion.

ab19569 at a 1/1000 diution staining VCAM1 in mouse brain tissue by Immunohistochemistry (frozen sections), incubated for 2 hours at 20°C. PFA fixed. Blocked with 10% serum for 1 hour at 20°C. Secondary used at 1/100 ab6733 Rabbit polyclonal to Rat IgG - H&L (Biotin).

DAB staining (brown colour). Counterstain is Chresyl violet.
