Our Abpromise guarantee covers the use of ab37985 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: Use a concentration of 0.3 - 1 µg/ml.Predicted molecular weight: 32 kDa.
ICC/IF: Use a concentration of 5 µg/ml
Forms a channel through the mitochondrial outer membrane that allows diffusion of small hydrophilic molecules. The channel adopts an open conformation at low or zero membrane potential and a closed conformation at potentials above 30-40 mV. The open state has a weak anion selectivity whereas the closed state is cation-selective.
Expressed in all tissues examined.
Belongs to the eukaryotic mitochondrial porin family.
Consists mainly of a membrane-spanning beta-barrel formed by 19 beta-strands.
Mitochondrion outer membrane.

Western blot - VDAC2 antibody (ab37985)

Anti-VDAC2 antibody (ab37985) at 1 µg/ml + Human Testis lysate (35µg protein in RIPA buffer).
Predicted band size : 32 kDa
Observed band size : 30 kDa (why is the actual band size different from the predicted?)
Primary incubation was 1 hour. Detected by
chemiluminescence.
Immunocytochemistry/ Immunofluorescence-VDAC2 antibody(ab37985)

ICC/IF image of ab37985 stained MCF7 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal donkey serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab37985, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 donkey anti-goat IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
