Thank you Karen for getting back to me. To make sure I am clear, you are saying that you no longer offer either the VDAC3 antibody (ab37459, raised in chickens) or VDAC1 (ab3434)?

ANSWER: |
Thank you so much for your patience. I have been in correspondence with the originator, and the protein the antibody was raised to was indeed not a chicken protein. However, I am not exactly clear which species it is from. The following reference describes the production of the antibody, and may be helpful for you: Mol. Gen.,Meta. 2000. 70:69-74. http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=10833333&dopt=Abstract (PubMed ID 10833333) In the article, they describe the immunization: "The histidine-tagged VDAC3 construct was created by PCR amplification of VDAC3 cDNA using a 3'65mer primer that incorporated the RGSH6 epitope at the 3' terminus of the PCR product. The tagged cDNA was T-A cloned to pBluescript, excised with ECoRI and NcoI, and cloned into the pET-28a expression vector. The VDAC3/pET construct was transformed into E. coli strain BL21, cells were grown to OD600, and protein expression was induced by the addition of 1mM IPTG. Cells were harvested 90 min. postinduction, and His-tagged proteins were purified on a nickel resin... Purity was verified by SDS-PAGE and protein conc. was determined by BCA assay. To produce antibodies, chickens were inoculated with 200 ug of VDAC3-His protein. Boost inoculations were performed every 3 weeks over a 12 week period with 100-200 ug pf protein, and serum reactivity was determined by ELISA. Antibodies were affinity-purified, where indicated, using the Aminolink Plus Kit.." Unfortunately, I do not have access to the journal this paper is published in, so I cannot take a look at the paper to see if I can find the species of the immunogen. However, it cannot be chicken, since there would be no immune response in chickens to a chicken protein. The originator also gave me a paper describing the antibody's use in Western blot and IHC: Arch Biochem Biophys; 2004. 422(2): 191-196. http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=14759607&dopt=Abstract (PubMed ID 14759607) I have added these references to the datasheet for future reference. I hope this information helps, please do not hesitate to contact us if you need any more advice or information. |
