Yes, I've looked up the Lab Vision Corporation and ordered another antibody for VEGF from them. If Vg76e can be used on native or denatured forms, then the antibody you supplied us with is not working. We've spend alot time trying to get it working and tried many experimental conditions, so you may want to note that for your records. You may also want to check the same batch or even my supply of it yourselves. |

Given the information that you have obtained it is highly plausible that the non-reducing conditions are necessary for the detection of VEGF using this clone. Is it necessary to use a non-denaturing gel or is it sufficient to leave out adding the reducing agent, like B-mercpatoethanol? Another question: do you know of any published literature which has used this antobody for western blotting. If so, could you send me a link to that? |

Yes, the omission of beta-mercaptoethanol would be sufficient for this kind of analysis. I will be back in touch when I have further information. |

I found some information from another company which said that this clone of antibody (VG76e) should only be used for un-reduced VEGF. Is it possible that this antibody only binds to an epitope that is exposed on the dimer? If so, could you suggest a protocol? Thank you and I look forward to hearing from you.

Thanks for your email. I'm replying to you on behalf of Chris, who is currently away on vacation. To use ab119 in IHC, I would recommend trying a range of dilutions in order to determine which is optimal for your particular experiment: 1:50, 1:100, 1:200. We do sell more of ab119 compared to ab3109. If you have any more questions, please let me know.
