Application notes

|
Recommended dilutions ELISA: Use at a concentration of 2.5 µg/ml. Allows the detection of 0.5- 1.0 ng/well of recombinant rat VEGF-C. IHC-P: Use at an assay dependent dilution (PMID 19412173). IP: Use at a concentration of 1 - 10 µg/ml IgG per 1ml lysate or reaction volume. WB: Use at a concentration of 1 µg/ml.
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Vascular endothelial growth factors (VEGFs), also known as vasculotropins, are a family of closely related growth factors having a conserved pattern of eight cysteine residues and sharing common VEGF receptors. VEGFs stimulate the proliferation of endothelial cells, induce angiogenesis, promote cell migration, increase vascular permeability, and inhibit apoptosis. The mitogenic activity of VEGFs appears to be mediated by specific VEGF receptors. The target cell specificity of VEGF is restricted to vascular endothelial cells. Vascular Endothelial Growth Factor C (VEGFC) is a member of the VEGF subfamily of PDGF-related growth factors. It is the ligand for Flt4 (VEGFR3) and KDR (VEGFR2). VEGFC binds Flt4 and induces tyrosine autophosphorylation of VEGFR3 and VEGFR2. VEGFC also stimulates the migration of bovine capillary endothelial cells in collagen gel. It is a specific growth factor for the lymphatic vascular system and mediates lymphangiogenesis. VEGFC is abundantly expressed in heart and skeletal muscle. Other tissues such as lung and kidney also express VEGFC.

Western blot - VEGFC antibody (ab9546)
Western blot of recombinant rat VEGF C from 2 samples of 15µl insect cell-conditioned media immobilized to PVDF-membrane.

Western blot - VEGFC antibody (ab9546)
Western blot with human and rat VEGF C and VEGF D using the polyclonal anti VEGF C antibody ab9546. Proteins were separated by 15% SDS-PAGE and blotted on to PVDF membranes as described. After saturation, the membrane was incubated for 1 h with ab9546 at 1 µg/ml vascular endothelial growth factor C in TBS containing 20% non-fat milk followed by incubation with a goat-anti-rabbit alkaline phosphatase-conjugated secondary antibody. Lane 1/250ng rat VEGF C; lane 2/250 ng human VEGF C; lane 3/250 ng rat VEGF D; lane4/250 ng human VEGF-D.
