Our Abpromise guarantee covers the use of ab68984 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
ICC/IF: 1/100
WB: 1/500Detects a band of approximately 72 kDa (predicted molecular weight: 59 kDa).(This antibody has been tested in Western blot against the recombinant peptide used as an immunogen. We have no data on detection of endogenous protein.)
IHC-P: 1/300 - 1/3000.Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
IHC-Fr: 1/300 - 1/3000.
Involved in acetylcholine transport into synaptic vesicles.
Peripheral and central cholinergic nervous systems.
Belongs to the major facilitator superfamily. Vesicular transporter family.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Vesicular Acetylcholine Transporter antibody (ab68984)

ab68984 at a 1/200 dilution staining Vesicular Acetylcholine Transporter in rat spinal cord tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Incubated overnight at 4°C, developed with DAB/Ni and counterstained with hematoxylin.
Immunocytochemistry/ Immunofluorescence - Anti-Vesicular Acetylcholine Transporter antibody (ab68984)

ICC/IF image of ab68984 stained PC12 cells. The cells were 4% formaldehyde (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab68984, 1/100 dilution) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
Western blot - Anti-Vesicular Acetylcholine Transporter antibody (ab68984)

All lanes : Anti-Vesicular Acetylcholine Transporter antibody (ab68984) at 1/500 dilution
Lane 1 : Spinal Cord (Mouse) Tissue Lysate
Lane 2 : Spinal Cord (Rat) Tissue Lysate
Lysates/proteins at 10 µg per lane.
Secondary
Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed (ab97080) at 1/5000 dilution
developed using the ECL technique
Performed under reducing conditions.
Predicted band size : 59 kDa
Observed band size : 72 kDa (why is the actual band size different from the predicted?)
Exposure time : 8 minutes
Vesicular Acetylcholine Transporter contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted.
ab68984 has not yet been referenced specifically in any publications.
