Our Abpromise guarantee covers the use of ab84888 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
IHC-P: 1/100 - 1/500.(Epitope exposure is recommended. Epitope exposure with citrate buffer will enhance staining.
Likely to work with frozen sections.)
ICC/IF: Use at an assay dependent dilution.
WDFY3 ( WD repeat and FYVE domain containing protein 3) was identified in a screen for phosphatidylinositol 3 phosphate [PtdIns(3)P] binding proteins. The FYVE domain is a zinc binding domain found in many PtdIns(3) binding proteins and is proposed to target proteins to membrane lipids. WDFY3 has been shown localized to the nuclear membrane and within autophagic membranes and it is proposed to function as a link between protein aggregates and the autophagic machinery. WDFY3 is also known as autophagy linked FYVE protein (Alfy).
Membrane; Peripheral membrane protein; Cytoplasmic side. Note: Colocalizes with autophagic structures in starved cells.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - WDFY3 antibody (ab84888)
Detection of Human WDFY3 in a formalin-fixed paraffin-embedded section of human ovarian carcinomaby by Immunohistochemistry using ab84888 at a dilution of 1:250. Detection: DAB staining.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - WDFY3 antibody (ab84888)
Detection of Human WDFY3 in a formalin-fixed paraffin-embedded section of human breast carcinomaby by Immunohistochemistry using ab84888 at a dilution of 1:250. Detection: DAB staining.
Immunocytochemistry/ Immunofluorescence - WDFY3 antibody (ab84888)

ab84888 staining WDFY3 in Human HeLa cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with formaldehyde, permeabilized with Triton X-100 0.5% in PBS and blocked with 2% BSA for 1 hour at 25°C. Samples were incubated with primary antibody (1/50 in PBS + 2% BSA) for 1 hour at 25°C. An Alexa Fluor®594-conjugated Donkey anti-rabbit monoclonal (1/2000) was used as the secondary antibody.
