Target

Relevance

WDFY3 ( WD repeat and FYVE domain containing protein 3) was identified in a screen for phosphatidylinositol 3 phosphate [PtdIns(3)P] binding proteins. The FYVE domain is a zinc binding domain found in many PtdIns(3) binding proteins and is proposed to target proteins to membrane lipids. WDFY3 has been shown localized to the nuclear membrane and within autophagic membranes and it is proposed to function as a link between protein aggregates and the autophagic machinery. WDFY3 is also known as autophagy linked FYVE protein (Alfy).

Cellular localization

Membrane; Peripheral membrane protein; Cytoplasmic side. Note: Colocalizes with autophagic structures in starved cells.

Cells were fixed with formaldehyde, permeabilized with Triton X-100 0.5% in PBS and blocked with 2% BSA for 1 hour at 25°C. Samples were incubated with primary antibody (1/50 in PBS + 2% BSA) for 1 hour at 25°C. An Alexa Fluor®594-conjugated Donkey anti-rabbit monoclonal (1/2000) was used as the secondary antibody.
