I have to contact you again, because of the antibody for XBP1 (ab37152). The problems with this antibody I explained you before. Consequently you sent me a new Aliquot of this antibody (Lot.No.: GR45274-2 ). I repeated my experiment, which I send you before, where I used the old aliquot of ab37152 I have (Lot.No.: 955532). Therefore see also the file attached. To summarize my results: The new antibody (Lot.No.: GR45274-2), which should be specific for both forms the unspliced and spliced XBP1, and the old antibody (Lot.No.: 955532 ) reveal the same reult by only detecting the spliced form of XBP1. Therefore I ask you to send me an aliquot of your u form specific antibody (ab37151) to exclude resp. verify the functionality of ab37152. Thanks a lot in advance |

To whom it may concern, I'm currently working on the protein XBP1, therefore I ordered your antibody ab37152, which should detect both forms of XBP1 (unspliced and spliced form). In different experimental approaches I tried to detect the unspliced form of XBP1 via wetsern blot. Therefore I overexpressed both the unspliced and the spliced form of XBP1 with respective expression plasmids in human cells. Additionally the degradation via proteasome and the splicing of the u-form by IRE1 were inhibited (Please see attachment). To summarize: I overexpressed the u-form of XBP1 and inhibited the conversion to the s-form and the degradation of the protein and could not detect the u-form with the XBP1 antibody ab37152. Therefore I would like to purchase the antibody only specific for the u-form of XBP ab37151. In the case that the antibody which should detect both forms, even detects the spliced form of XBP1, I would be very pleased, if you could give me an appropriate offer for this antibody. It would be also possible to send me a small aliquot of the antibody (u-specific, ab37151) to test it for my application. About a further troobleshooting concerning the detection of the unspliced form with antibody ab37152 I would be very pleased as well. Immunocytochemistry of XBP-1 in HepG2 cells with ab37152 at 10 µg/ml
ab37152 at 1/200 dilution staining XBP1 in human non-lesional skin tissue sections by Immunohistochemistry (Formalin/ PFA fixed paraformaldehyde tissue sections). A biotin labelled secondary was used at 1/100 dilution. Diaminobenzidine (DAB) was used for staining development in a ABC staining Kit and the sections were counterstained with haematoxylin.

ab37152 staining XBP1 in human liver tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue was fixed with formaldehyde and a heat mediated antigen retrieval step was performed using Citrate-EDTA antigen retrieval buffer and microwave. Heating was performed in the microwave 3 minutes on high until boiling, 6 minutes on 20% power. Samples were then blocked for 15 minutes at 27ºC followed by incubation with the primary antibody at a 1/100 dilution for 12 hours at 4ºC. An undiluted HRP-conjugated rat polyclonal was used as the secondary antibody.
