Recommended dilutions ICC/IF: Use at a concentration of 10 µg/ml. IHC-P: Use at an assay dependent dilution (PMID 19543371). WB: Use at a concentration of 1 - 2 µg/ml. Predicted molecular weight: 29 kDa (isoform 1) and 40 kD (isoform 2). Can be blocked with XBP1 peptide (ab39910).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
This gene encodes a transcription factor that regulates MHC class II genes by binding to a promoter element referred to as an X box. The protein contains two domains commonly found in transcription factors that confer DNA binding and dimerization capability. This gene product was also separately identified as a cellular transcription factor that binds to an enhancer in the promoter of the T cell leukemia virus type 1 promoter. It may increase expression of viral proteins by acting as the DNA binding partner of a viral transactivator. There is also a recent finding that the same agents that cause an ER stress response, the unfolded protein response, also increase transcription of this gene. This increase in transcription requires an ER stress response consensus binding element in the promoter.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - XBP1 antibody (ab37152)
ab37152 at 1/200 dilution staining XBP1 in human non-lesional skin tissue sections by Immunohistochemistry (Formalin/ PFA fixed paraformaldehyde tissue sections). A biotin labelled secondary was used at 1/100 dilution. Diaminobenzidine (DAB) was used for staining development in a ABC staining Kit and the sections were counterstained with haematoxylin.
