Recommended dilutions ICC: 1/200 - 1/500. IP: Use at a concentration of 2 - 6 µg/ml. WB: 1/1000 - 1/10000. Detects a band of approximately 90 kDa (predicted molecular weight: 70 kDa).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

XRCC1 is involved in the efficient repair of DNA single-strand breaks formed by exposure to ionizing radiation and alkylating agents. This protein interacts with DNA ligase III, polymerase beta and poly (ADP-ribose) polymerase to participate in the base excision repair pathway. It may play a role in DNA processing during meiogenesis and recombination in germ cells. A rare microsatellite polymorphism in this gene is associated with cancer in patients of varying radiosensitivity. In testis XRCC1 is expressed at high levels.

All lanes : XRCC1 antibody (ab9147) at 1 µg/ml
Lane 1 : Whole cell extract (50mcg) from mock treated HeLa cells. Lane 2 : Whole cell extract (50mcg) from siRNA treated HeLa cells.
Predicted band size : 70 kDa Observed band size : 76,90 kDa (why is the actual band size different from the predicted?)
Detection: ECL.

Immunocytochemistry/ Immunofluorescence - XRCC1 antibody (ab9147)
ab9147 staining human HeLa cells by ICC/IF. Cells were PFA fixed and permeabilized in 0.5% Triton X100 prior to blocking in 5% BSA for 30 minutes at 20°C. The primary antibody was diluted 1/200 and incubated with the sample for 1 hour at 20°C. A Cy3® conjugated donkey anti-rabbit antibody diluted 1/400 was used as the secondary.
