Recommended dilutions
WB: 1/500 - 1/1000. Detects a band of approximately 63 kDa (predicted molecular weight: 49.7 kDa). For optimal results, primary antibody incubations should be performed at 4°C.
ICC/IF: Use at a concentration of 1 - 5 µg/ml.

Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance 

YY1 is a ubiquitously distributed transcription factor belonging to the GLI Kruppel class of zinc finger proteins. The protein is involved in repressing and activating a diverse number of promoters. YY1 may direct histone deacetylases and histone acetyltransferases to a promoter in order to activate or repress the promoter, thus implicating histone modification in the function of YY1.

Detection of YY1 by Western blot analysis. YY1 can be detected in nuclear extracts derived from Raji cells using ab12132 (lane 1) at a 1:1000 dilution. The signal corresponding to nuclear YY1 is eliminated by adding the immunizing peptide to the reaction mixture (lane 2).

ICC/IF image of ab12132 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab12132, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
