Overview
Fluorescent 5-HT1B antagonist
Excitation wavelength: 633 nm
Emission wavelength: 650 nm
Properties
To determine the apparent KD for ab118650, cells were treated with varying concentrations of 5-CT or CP-94253 alone, or in the presence of 1 µM ab118650, and the cyclic-AMP induced expression of SPAP measured.
The apparent KD was calculated from the rightward shift of the agonist response curve in the presence of ab118650, compared to the response curve for the agonist alone.

ab118650 ligand binding and displacement. Left: The ab118650 ligand (100 nM) binding to CHO cells expressing 5-HT1B receptors. Center: Binding in the same 5-HT1B expressing cell line blocked by the unlabelled competitor CP-94253 at 1 µM. Right: Lack of strong binding of ab118650 to CHO cells expressing 5-HT1A receptors. Nuclei have been counter-stained with Hoechst.

Similar studies were also carried out using a CHO cell line expressing the Human 5-HT1A receptor to test for receptor selectivity. ab118650 did not show either agonist or antagonist activity at the Human 5-HT1A receptor. Antagonist –log KD values for 5-HT1B 6.71 (vs 5-CT)and 6.86 (vs CP-94253)
