Application notes
(see key)

Recommended dilutions
ICC/IF: Use at a concentration of 5 µg/ml.
WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 50 kDa (predicted molecular weight: 30 kDa).

Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Research areas

Chromatin and Nuclear Signaling >> Transcription >> Other factors

Relevance

Zhangfei strongly activates transcription when bound to HCFC1. It suppresses the expression of HSV proteins in cells infected with the virus in a HCFC1-dependent manner. It also suppresses the HCFC1 dependent transcriptional activation by CREB3 and reduces the amount of CREB3 in the cell. It can down-regulate expression of some cellular genes in ZF expressing cells.

Secondary
IRDye 680 Conjugated Goat Anti-Rabbit IgG (H+L) at 1/10000 dilution

Performed under reducing conditions.

Predicted band size : 30 kDa
Observed band size : 50 kDa (why is the actual band size different from the predicted?)

We are unsure about the expression of the Zhangfei protein so cannot explain the bands observed in Western Blot.  Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.

Immunocytochemistry/ Immunofluorescence - Zhangfei antibody (ab28700)

ICC/IF image of ab28700 stained human HEK293 cells. The cells were PFA fixed (10 min) and incubated with the antibody (ab28700, 5µg/ml) for 1h at room temperature. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Image-iTTM FX Signal Enhancer was used to quench autofluorescence.  5% BSA (in TBS-T) was used for all other blocking steps. DAPI was used to stain the cell nuclei (blue). Alexa Fluor® 594 WGA was used to label plasma membranes (red).
