ab123457 SIRT1 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human SIRT1 in cell and tissue lysates. The assay employs an antibody specific for Human SIRT1 protein coated onto well plate strips.
Standards and samples are pipetted into the wells and analyte present in the sample is bound to the wells by the immobilized antibody. The wells are washed and an anti- SIRT1 primary detector antibody is added. After washing away unbound primary detector antibody, HRP-conjugated secondary detector antibody specific for the primary detector antibody is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of analyte bound. The developing blue color is measured at 600 nm.
Optionally the reaction can be stopped by adding hydrochloric acid which changes the color from blue to yellow and the intensity can be measured at 450 nm.
Species– human. Rat, mouse are not suitable with this kit, other species are untested.

Whole cultured cell extract: 0.060 - 1 mg/mL
Human SIRT1 standard: 0.008 - 0.5 µg/mL
Store all components at 4°C. This kit is stable for at least 6 months from receipt. After reconstitution the standard should be stored at -80°C. Unused microplate strips should be returned to the pouch containing the desiccant and resealed.
