Target
- FunctionHistone H3-like variant which exclusively replaces conventional H3 in the nucleosome core of centromeric chromatin at the inner plate of the kinetochore. Required for recruitment and assembly of kinetochore proteins, mitotic progression and chromosome segregation. May serve as an epigenetic mark that propagates centromere identity through replication and cell division.

Anti-CENPA antibody images

ab20665 at a 1/200 dilution staining paraformaldehyde fixed asynchronous HeLa cells by ICC/IF. The cells were incubated with the antibody overnight and then bound antibody was detected using a Cy3 conjugated goat anti-rabbit antibody. This antibody localizes to centromeres as identified by ACA staining (not shown). Nuclei were visualised with DAPI staining.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-CENPA antibody (ab20665)IHC image of CENPA staining in Human breast adenocarcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab20665, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
