Target
- FunctionStacking factor involved in the postmitotic assembly of Golgi stacks from mitotic Golgi fragments. Key structural protein required for the maintenance of the Golgi apparatus integrity: its caspase-mediated cleavage is required for fragmentation of the Golgi during apoptosis. Also mediates, via its interaction with GM130, the docking of transport vesicles with the Golgi membranes.

modificationsPhosphorylated by CDC2/B1 and PLK kinases during mitosis. Phosphorylation cycle correlates with the cisternal stacking cycle. Phosphorylation of the homodimer prevents the association of dimers into higher order oligomers, leading to cisternal unstacking.
Target for caspase-3 cleavage during apoptosis. The cleavage contributes to Golgi fragmentation and occurs very early in the execution phase of apoptosis. - Cellular localizationGolgi apparatus > cis-Golgi network membrane. Undergoes rapid exchange with the cytosol.

Immunocytochemistry/ Immunofluorescence - GRASP65 antibody (ab30315)This image is courtesy of an Abreview submitted by Mr Carl HobbsGRASP65 antibody (ab30315) labelling in mouse Cor1 cell cells. Cor1 cells were fixed with formaldehyde and permeabilised. 1% BSA used as blocking agent for 10 mins. Primary antibody ab30315 incubated at rt°C at 1/5000 for 2 hours in TBS/BSA/azide/Triton. Secondary Antibody Name: anti rabbit IgG Alexa Fluor® 594 1/1000. ab30315 in Cor1 cells appears to localise discretely to an intracellular compartment that could be the Golgi Appparatus. Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - GRASP65 antibody (ab30315)Image courtesy of an Abreview submitted by Carl Hobbs.ab30315 staining GRASP65 in rat hippocampal tissue (sub-region) by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue was fixed with formaldehyde and a heat mediated antigen retrieval step was performed using citric acid pH 6.0. Samples were then blocked with 1% BSA for 10 minutes at room temperature followed by incubation with the primary antibody at a 1/6000 dilution for 16 hours. A biotin-conjugated anti-rabbit was used as secondary antibody at a 1/300 dilution. Nuclei are lightly counterstained with Haemalum. -
