    Predicted to work with: Human
  • Immunogen

Synthetic Peptide: A(5)NNSVAYSGVKNSIKEAN(22). Immunizing peptide corresponds to amino acid residues 5-22 from rat PMCA 1 ATPase protein. This sequence is completely conserved between rat and human PMCA 1 ATPase.

Properties

Applications

Our Abpromise guarantee covers the use of ab3528 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Notes
ICC/IF ICC/IF: 1/300.
ELISA ELISA: Use at an assay dependent dilution.
WB WB: 1/1000. This antibody detects ~134 kDa and ~130 kDa proteins representing PMCA 1a and 1b ATPase from rat samples.
IHC-P IHC-P: Use at an assay dependent dilution.

Target

  • FunctionThis magnesium-dependent enzyme catalyzes the hydrolysis of ATP coupled with the transport of calcium out of the cell.
  • Tissue specificityIsoform B is ubiquitously expressed. Isoform C is found in brain cortex, skeletal muscle and heart muscle. Isoform D has only been found in fetal skeletal muscle. Isoform K has been found in small intestine and liver.
  • Sequence similaritiesBelongs to the cation transport ATPase (P-type) (TC 3.A.3) family. Type IIB subfamily.
  • DomainThe calmodulin-binding subdomain B is different in the different splice variants and shows pH dependent calmodulin binding properties in isoforms A, C, D and E.
  • Cellular localizationCell membrane.

Anti-PMCA1 antibody images

  • ab3528 at 1/100 staining rat brain hippocacmpal slices and slice cultures by IHC-P. The tissue was paraformaldehyde fixed, permeabilized with Triton X-100 and blocked with BSA before incubation with the antibody for 12 hours. An Alexa Fluor ® 488 conjugated goat anti-rabbit antibody was used as the secondary.

  • developed using the ECL technique

    Performed under reducing conditions.

Observed band size : 123 kDa (why is the actual band size different from the predicted?)

    Exposure time : 2 minutes

  • Immunohistochemistry was performed on normal biopsies of deparaffinized Human skeletal muscle tissue. To expose target proteins heat induced antigen retrieval was performed using 10mM sodium citrate (pH6.0) buffer microwaved for 8-15 minutes. Following antigen retrieval tissues were blocked in 3% BSA-PBS for 30 minutes at room temperature. Tissues were then probed at a dilution of 1:20 with a rabbit polyclonal antibody recognizing PMCA1 ATPase ab3528 or without primary antibody (negative control) overnight at 4°C in a humidified chamber. Tissues were washed extensively with PBST and endogenous peroxidase activity was quenched with a peroxidase suppressor. Detection was performed using a biotin-conjugated secondary antibody and SA-HRP followed by colorimetric detection using DAB. Tissues were counterstained with hematoxylin and prepped for mounting.
