This antibody recognizes the Propeptide region of MMP13. The Propeptide region is shed during enzymatic activation of MMP13, and the antibody does not recognize the activated forms. The antibody can thus be used to discriminate between the zymogen and activated MMP13.

Recommended dilutions
WB: Recommended starting dilution of 1/3000 (when using colorimetric substrates such as BCIP/NBT) and 1/6000 (for chemiluminescent substrates). Higher concentrations of antibody may be needed for non human samples. The antibody recognizes the latent proenzyme, at 60 kDa, as well as the active form at 48 kDa, and intermediate activation forms. When used against the reduced protein, the antibody identifies a band at 60 kDa (predicted molecular weight: 54 kDa). Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance 

Collagenase 3 (MMP13) is a secreted 452 residue protein which is released from cells as aninactive zymogen and activated extracellularly by removal of its propeptide. MMP13 was first identified in tumor cells but has since been found in synoviocytes and normal fibroblasts stimulated by IL6 or a combination of TNF alpha and IL1. Under normal conditions, MMP13 is expressed during embryogenesis (fetal bone development) and is present only at very low levels in adult tissue. However, this enzyme is reported to be involved in the development and metastasis of breast and lung carcinomas, chondrosarcomas and osteosarcomas, head and neck carcinomas and some forms of skin cancer. Additionally, this enzyme plays an important role in degenerative bone diseases including osteoarthritis and rheumatoid arthritis. In rodents, MMP13 is thought to fill the role of MMP1, which they appear to lack.

The low endogenous MMP13 protein levels may require concentration of samples by ultrafiltration or ammonium sulfate precipitation prior to Western blotting.
