Specificity 

ab39029 recognises the Carboxyterminal end of MMP19. It does not recognise the smallest active form of MMP19 as the hemopexin domain is cleaved from it.
This antibody will bind to the reduced and non reduced form of MMP19, but does not cross react with the other MMP family members (MMP1, MMP2, MMP3, MMP9, etc.)

Recommended dilutions
WB: 1/1000 when using colorimetric substrates such as BCIP/NBT - 1/5000 when using chemiluminescent substrates. Predicted molecular weight: 57 kDa. When used against the reduced protein, ab39029 identifies bands at 58 Kd, 45 Kd, and a series of further cleaved active forms. Note: the conditioned media may need to be concentrated for Western blot. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance 

Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development,reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. MMP19 is one of the more recently identified MMPs, has several novel structural features, and so does not fall into the traditional MMP groupings. It is without a furin cleavage site or transmembrane domain and unlike the other MMPs, MMP19 has an 8-residue acidic patch at the beginning of the hinge region. MMP19 is not constituitively produced by most cell types; it has been found in lung fibroblasts, and gingival fibroblasts when stimulated with TPA, as well as breast tumor and osteosarcoma cell lines. MMP19 may play a role in pathological processes participating in rheumatoid arthritis (RA)-associated joint tissue destruction. Autoantigens to anti-MMP19 are frequent in RA patients.
