| Application notes (see key) | Recommended dilutions WB: Recommended starting dilution of 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non human samples. Detects bands of approximately 70kDa and 66kDa when used against the reduced protein. It is unclear if this represents alternative splicing, post translational modifications, aggregation, or some other phenomenon. (Predicted molecular weight: 53 kDa). | Relevance | Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development,reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMPs are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The enzyme encoded by this gene lacks the signal sequence found in other MMPs, suggesting that it may function intracellularly. The substrate for the enzyme has not been identified, but it is thought that the protein may play a role in reproductive processes. This protein is encoded by the MMP22 or MMP23B gene which is part of a duplicated region of chromosome 1p36.3, which includes a second gene, MMP23A, which is almost identical.
MMP22 was first cloned from chicken fibroblasts, and named CMMP. Sequence comparisons confirmed CMMP was a metalloproteinase, but homology between chicken and human MMPs was too low to assign CMMP as a human ortholog. Several years later CMMP was given the moniker MMP22. A BLAST of chicken MMP22 finds human MMP27 as the closest human sequence. Chicken MMP22 compares most closely to human MMP27, with 59% identity (compared to 51% identity between CMMP and human MMP3). It is hazardous to guess the identity of species orthologs between chicken and human proteins: chicken MMP2 is 84% identical to human and chicken MMP13 is 75% identical to human, but chicken MMP9 is only 59% identical to human MMP9. The chicken MMP22 sequence encodes a 472 amino acid protein, with predicted molecular weight of 53.254 kD. Although MMP22 was thought to be most like human MMP3 (Stromelysin 1), chicken MMP22 protein has a quite basic predicted isoelectric point of 9.104, while human MMP3 has a predicted isoelectric point of 5.699. Human MMP27 has a predicted isoelectric point of 8.826, making it similar to CMMP. The signal sequence, and lack of a transmembrane domain lead to the assumption that MMP22 is a secreted MMP, and it is tempting to speculate that the basic isoelectric point might direct the MMP22 to interact with ECM. It is still uncertain if MMP22 is a unique MMP, or the chicken ortholog of human MMP27. |
