| Application notes (see key) | Recommended dilutions WB: 1/1000 when using colorimetric substrates such as BCIP/NBT - 1/5000 for chemiluminescent substrates. Detects bands of approximately 53 and 51 kDa (the pro form), as well as the smaller activated forms. (Predicted molecular weight: 57 kDa). Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development,reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. MMP19 is one of the more recently identified MMPs, has several novel structural features, and so does not fall into the traditional MMP groupings. It is without a furin cleavage site or transmembrane domain and unlike the other MMPs, MMP19 has an 8-residue acidic patch at the beginning of the hinge region. MMP19 is not constituitively produced by most cell types; it has been found in lung fibroblasts, and gingival fibroblasts when stimulated with TPA, as well as breast tumor and osteosarcoma cell lines. MMP19 may play a role in pathological processes participating in rheumatoid arthritis (RA)-associated joint tissue destruction. Autoantigens to anti-MMP19 are frequent in RA patients. |
