Ab43868 at 1/10000 dilution staining rat tissue; positive neurons in rat brainstem, vibratome sectioned, ABC / DAB detection.

ICC/IF image of ab43868 stained PC12 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab43868, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Samples were then permeabilized using 0.1% Tween in TBS, blocked with 10% serum for 1 hour at 23°C and then incubated with ab43868 at a 1/200 dilution for 12 hours at 4°C. The secondary used was a biotinylated goat anti-rabbit IgG (H+L) used at a 1/200 dilution. -
Electron Microscopy - Anti-Dopamine beta Hydroxylase antibody (ab43868)This image is courtesy of an anonymous Abreviewab43868 was used to detect Dopamine beta Hydroxylase in rat brain tissue for Electron Microscopy (EM). The Tissue was Perfusion fixed with 4% PFA and the stored for less than 1 week in 1% PFA refrigerated, tissue blocked with PBS-BSA for 15 min, incubation was 1 hour with primary antibody at room temperature. A 10nm Gold-conjugated antirabbite polyclonal(1/20) was used a secondary antibody.
