Target

  • RelevanceCadherins are a family of transmembrane glycoproteins that play a key role in Calcium dependent cell-cell adhesion. Several members of the cadherin family have been identified so far, including E- (epithelial), P- (placental), N- (neuronal) and M- (muscle) cadherin. Cadherins consist of an extracellular domain containing 5 cadherin domains, a transmembrane region, and a conserved cytoplasmic domain. Transcripts from this particular cadherin are expressed in myoblasts and upregulated in myotubule-forming cells. M-Cadherin is thought to be essential for the control of morphogenetic processes, specifically myogenesis, and may provide a trigger for terminal muscle cell differentiation.

  • Anti-M Cadherin [12G4] antibody (ab78090) at 0.5 µg/ml + Lysates of C2C12 cells differentiated into myotubes

    Secondary
    Goat anti-mouse HRP conjugate
    developed using the ECL technique

Predicted band size : 90 kDa
Observed band size : 130 kDa (why is the actual band size different from the predicted?)
  • ab78090 staining M Cadherin in C2C12 murine myoblasts (myotubes) by Immunocytochemistry/ Immunofluorescence. The cells were fixed in paraformaldehyde, permeabilised in 0.1% Triton and then blocked using 0.2% BSA, 1% goat serum for 2 hours at 25°C. Samples were then incubated with primary antibody at 1/100 for 12 hours at 4°C. The secondary antibody used was a goat anti-mouse IgG conjugated to Alexa Fluor® 488 (green) used at a 1/200 dilution.
