Applications
Our Abpromise guarantee covers the use of ab9704 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
| Application | Notes |
|---|---|
| WB | WB: Use at an assay dependent concentration. To detect hPDGF-BB by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hPDGF-BB is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions. |
| ELISA | ELISA: Use at an assay dependent concentration. To detect hPDGF-BB by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hPDGF-BB. |
| Neutralising | Neut: Use at an assay dependent concentration. To yield one-half maximal inhibition [ND50] of the biological activity of hPDGF-BB (2.5 ng/ml), a concentration of 0.03 - 0.04 µg/ml of this antibody is required. |
| Sandwich ELISA | sELISA: Use at an assay dependent concentration. To be used with ab84265 as detection antibody and ab9706 as protein standard. |
| IHC-P | IHC-P: Use a concentration of 1 µg/ml. |

ab9704 staining PDGF BB in normal human skin tissue section by Immunohistochemistry (Formalin/PFA fixed paraffin-embedded sections). Tissue underwent proteinase K mediated antigen retrieval. The primary antibody was used at 1 ug/ml and incubated with sample at 4°C overnight. A HRP-labeled polymer detection system was used with a non-alcohol soluble AEC chromogen.
