Target

  • Function
    Iron sensor. Binds a 4Fe-4S cluster and functions as aconitase when cellular iron levels are high. Functions as mRNA binding protein that regulates uptake, sequestration and utilization of iron when cellular iron levels are low. Binds to iron-responsive elements (IRES) in target mRNA species when iron levels are low. Binding of a 4Fe-4S cluster precludes RNA binding.
    Catalyzes the isomerization of citrate to isocitrate via cis-aconitate.

  • Immunofluorescence staining of HepG2 cells with purified ab126595 at a working dilution of 1 in 250, counter-stained with DAPI. The secondary antibody was Alexa Fluor® 488 goat anti rabbit, used at a dilution of 1 in 500. The cells were fixed in 4% PFA. The negative control is shown in bottom right hand panel - for the negative control, purified ab126595 was used at a dilution of 1/250 followed by an Alexa Fluor® 488 goat anti-mouse antibody at a dilution of 1/500.

  • Flow Cytometry analysis of HeLa (human cervix adenocarcinoma) cells labeling Aconitase 1 with purified ab126595 at 1/130 dilution (10ug/ml) (red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. A Goat anti rabbit IgG (Alexa Fluor® 488)(1/2000 dilution) was used as the secondary antibody. Rabbit monoclonal IgG (Black) was used as the isotype control, cells without incubation with primary antibody and secondary antibody (Blue) was used as the unlabeled control.
