| Application notes (see key) | Recommended dilutions ICC/IF: Use at a concentration of 10 µg/ml. WB: 1/1000 when using colorimetric substrates such as BCIP/NBT, and 1/5000 for chemiluminescent substrates. Detects a band of approximately 98 kDa due to glycosylation and the cyteine-rich regions and 58-60 when Furin processed. (predicted molecular weight: 84 kDa). EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. Dilution optimised using Chromogenic detection.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | ADAM10 is a member of the ADAM (a disintegrin and metalloprotease like domain) family. It cleaves the membrane-bound precursor of TNF-alpha at 76-Ala-|-Val-77 to its mature soluble form and is responsible for the proteolytic release of several cell-surface proteins, including heparin binding epidermal growth-like factor, ephrin-A2 and for constitutive and regulated alpha-secretase cleavage of amyloid precursor protein (APP). It contributes to the normal cleavage of the cellular prion protein and is involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicules, suggesting a vesicle-based protease activity. |
