Specificity
ab38981 recognises ADAM8 at the Carboxiterminal domain. It does not react with other ADAMs.
We have a range of domain specific antibodies for this target. For a full list please see all ADAM8 antibodies

Application notes

Recommended dilutions WB: 1/1000 when using colorimetric substrates such as BCIP/NBT - 1/5000 when using chemiluminescent substrates. Predicted molecular weight: 88 kDa. But note that ab38981 recognizes a 70 kD band in reduced Western blots of conditioned media or cell lysates. Also note that EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
Members of ADAM family are cell surface proteins with a unique structure possessing both potential adhesion and protease domains. The extracellular region of ADAM8 shows significant amino acid sequence homology to hemorrhagic snake venom proteins, including the metalloprotease and disintegrin domains. The expression of ADAM8 is upregulated by retinoic acid and vitamin D3. The function of ADAM8 is still poorly understood. ADAM8 has been shown to stimulate osteoclasts, and to be produced by bone marrow cultures stimulated by vitamin-D. ADAM8 is involved in protein processing at the cell surface and is also thought to act as a leukocyte differentiation marker.
