Application notes

WB: 1/1,000 when using colorimetric substrates such as BCIP/NBT and 1/5,000 for chemiluminescent substrates. Predicted molecular weight: 184 kDa. This antibody recognizes the zymogen of ADAMTS7 at around 300 kD in reduced Western blots, activated or alternatively spliced forms at 180-120 kD (major bands) and breakdown products at 58-45 kD in cell lysates. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance

ADAMTS proteases are secreted enzymes containing a prometalloprotease domain of the reprolysin type. The ADAMTS proteases function in processing of procollagens and von Willebrand factor as well as catabolism of aggrecan, versican and brevican. They have been demonstrated to have important roles in connective tissue organization, coagulation, inflammation, arthritis, angiogenesis and cell migration. A member of the metalloproteinase family containing disintegrin like domains (ADAMs), the function of ADAMTS7 is still poorly understood. ADAMTS7 most resembles ADAMTS12 in overall structure. ADAMTS7 does not have a transmembrane domain, unlike many of the ADAMs proteases, and is a secreted protein.
