I purchased antibodies for detecting AFP in serum by ELISA assay. Details of my antibodies are: 1. alpha 1 Fetoprotein antibody (ab 10071) with lot number #156917 2. alpha 1 fetoprotein antibody (ab 10072) with lot number #159812 Those antibodies will be applied as capture (no 1) and detecting (no 2) antibodies. What I want to know is what dilution should I perform for those antibodies to optimize my ELISA assay in measuring AFP level in serum? Thanks for your help |

Thank you for your enquiry. Unless a dilution is listed on the data sheet, we do not know the dilution to use for sandwich ELISA. A few things to keep in mind are that typically you do not need a lot of antibody to do an ELISA, just enough to coat the wells. Perhaps you could start with 0.1 ug/mL. ELISAs can be tricky because there is a bit of trial and error involved if you don't have a recommended dilution. It may take some time to optimize your assay. Maybe you could try a few dilutions between 0.1 ug/ml and 5 ug/ml and see how that goes. I am sorry that I do not have more information.
