    Alpha 2 Macroglobulin is a major serum protein with diverse functions, including inhibition of protease activity and binding of growth factors, cytokines, and disease factors. Increased serum alpha-2-Macroglobulin has been suggested to be associated with multiple sclerosis (MS), glomerular disease, and with liver diseases.

    ab108888 is designed for detection of human alpha 2 Macroglobulin in plasma, and serum. This assay employs a quantitative competitive enzyme immunoassay technique that measures human alpha 2 Macroglobulin in less than 3 hours. A polyclonal antibody specific for human alpha 2 Macroglobulin has been pre-coated onto a 96-well microplate with removable strips. Alpha 2 Macroglobulin in standards and samples is competed by a biotinylated alpha 2 Macroglobulin sandwiched by the immobilized antibody and streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.

Properties

  • FunctionIs able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase.
