The immunogen region (90-120) appears to refold and form non-native disulphide bonds after the protein has been transferred to nitrocellulose and left in biological buffer for longer than 24 hrs before applying ab12271. This refolding prevents ab12271 from accessing its epitope. Hence it is important to reduce and alkylate samples prior to SDS-PAGE and immunoblotting. It is possible to reduce and alkylate the proteins after transfer to nitrocellulose and before immunoblotting with ab12271.|
