Target

  • FunctionConverts angiotensin I to angiotensin II by release of the terminal His-Leu, this results in an increase of the vasoconstrictor activity of angiotensin. Also able to inactivate bradykinin, a potent vasodilator. Has also a glycosidase activity which releases GPI-anchored proteins from the membrane by cleaving the mannose linkage in the GPI moiety.
  • Tissue specificityUbiquitously expressed, with highest levels in lung, kidney, heart, gastrointestinal system and prostate. Isoform Testis-specific is expressed in spermatocytes and adult testis.
  • Involvement in diseaseIschemic stroke (ISCHSTR) [MIM:601367]: A stroke is an acute neurologic event leading to death of neural tissue of the brain and resulting in loss of motor, sensory and/or cognitive function. Ischemic strokes, resulting from vascular occlusion, is considered to be a highly complex disease consisting of a group of heterogeneous disorders with multiple genetic and environmental risk factors. Anti-Angiotensin Converting Enzyme 1 antibody [9B9] images

  • ab77990, at a 1/100 dilution, staining Angiotensin Converting Enzyme 1 in rat heart myocardium (three weeks after a myocardial infarction) by Immunohistochemistry.
  • ab77990, at a 1/100 dilution, staining Angiotensin Converting Enzyme 1 in human kidney by Immunohistochemistry.
  • Overlay histogram showing THP1 cells stained with ab77990 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab77990, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive result in 80% methanol (5 min) fixed THP1 cells used under the same conditions.

We don't have any specific recommendations for western blotting protocol for ab32212. Although a good starting dilution for western blotting using ab32212 is noted as 1:5000.

For ab11734 I'd recommend starting with a dilution of 1: 250 and dilute ab87436 to 1:1000. The primary antibody diluent could be TBS with 0.05% Tween 20.
