A major contributor to cellular homeostasis is the ability of the cell to strike a balance between the formation and degradation/removal of its cellular components. This process of internal cellular turn-over is called autophagy (self-eating), and is facilitated by a pathway of around 16 interacting proteins in the human. APG3 is a conjugating enzyme for APG8 lipidation, an essential step for the initiation of autophagy. APG16 is essential for autophagy and interacts with ATG5 as part of either the minor or major complexes, respectively composed of 4 sets of ATG12-ATG5 and APG16 (400 kDa) or 8 sets of ATG12-ATG5 and ATG16 (800 kDa). A disruption to the autophagic process is now associated with the progression of several cancers, neurodegenerative disorders and cardiac pathologies.

Note: Multiple isoforms of agp16 can be detected in mouse skeletal muscle lysate using ab72086. The approximately 40 kD band seen in this lysate is also seen in prebleed and is not believed to be relevant.
