Our Abpromise guarantee covers the use of ab85656 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 µg/ml. Predicted molecular weight: 45 kDa.Can be blocked with Arg 3.1 peptide (ab117435). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 1 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

  • FunctionRequired for consolidation of synaptic plasticity as well as formation of long-term memory. Regulates endocytosis of AMPA receptors in response to synaptic activity. Required for homeostatic synaptic scaling of AMPA receptors (By similarity). Plays a role in the regulation of cell morphology and cytoskeletal organization. Required in the stress fiber dynamics and cell migration.
  • Sequence similaritiesBelongs to the ARC/ARG3.1 family.
  • Cellular localizationCytoplasm, cytoskeleton. Endosome. Cytoplasmic vesicle, secretory vesicle, acrosome. Cell junction, synapse, postsynaptic cell membrane, postsynaptic density. Cell projection, dendrite. Cell projection, dendritic spine. Cell junction, synapse. Associated with the cell cortex of neuronal soma and dendrites. Enriched in postsynaptic density of dendritic spines. Associated with the sperm tail (By similarity). Enriched on the plasma membrane.

  • IHC image of ab85656 staining in human hippocampus formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab85656, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human adrenal tissue labelling Arg 3.1 with ab85656 at 5µg/ml.
