Applications

Our Abpromise guarantee covers the use of ab166850 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 µg/ml. Detects a band of approximately 240 kDa (predicted molecular weight: 197 kDa).

Target

  • Function
Involved in transcriptional activation and repression of select genes by chromatin remodeling (alteration of DNA-nucleosome topology). Required for the stability of the SWI/SNF chromatin remodeling complex SWI/SNF-B (PBAF). May be involved in targeting the complex to different genes. May be involved in regulating transcriptional activation of cardiac genes.
  • Tissue specificity
Highly expressed in heart and testis.
  • Sequence similarities
    Contains 1 ARID domain.
    Contains 1 C2H2-type zinc finger.
    Contains 1 RFX-type winged-helix DNA-binding domain.
  • Developmental stage
Up-regulated in adult heart (at protein level).
  • Cellular localization
    Nucleus.

  • Anti-ARID2 antibody (ab166850) at 1 µg/ml (Milk blocking 3%) + Human testis tissue lysate - total protein (ab30257) at 20 µg

    Secondary
    Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/50000 dilution

    Developed using the ECL technique.

    Performed under reducing conditions.

    Predicted band size: 197 kDa
Observed band size: 240 kDa (why is the actual band size different from the predicted?)

    Exposure time: 20 minutes

This blot was produced using a 3-8% Tris Acetate gel under the TA buffer system. The gel was run at 150V for 60 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab166850 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406.
