Question 1

Tuesday 01-May-2012

We plan on trying out one of your suggested anti-beta 2 microglobulin antibody (ab75853) first and participate in the testing discount program.   I just want to verify if there is any code associated with the program (is it the alphanumeric one in the brackets in the subject heading of the email?) and whether we need to notify you about which the products we plan to test in advance.  We will also most likely test the suggested ab99977 ELISA kit at some point but for now will order just the antibody listed above.

ANSWER:

Thank you for your response and for participating in our Abreview Testing program. The code in brackets in the subject heading is a reference number for these correspondences. Should you wish to test another of the products, referencing this number would be very helpful.

I am very pleased to hear you would like to accept our offer and test ab75853 in porcine tissues. This code will give you: 1 free PRIMARY ANTIBODY OR VALUE OFF OF ORDER before the expiration date. To redeem this offer, please submit an Abreview for Porcine and include this code in the “Additional Comments” section so we know the Abreview is for this promotion. For more information on how to submit an Abreview, please visit the site: www.abcam.com/Abreviews.

Remember, we publish both positive and negative Abreviews on our datasheets so please submit the results of your tests. The code will be active once the Abreview has been submitted and can be redeemed in one of the following ways: 1) Call to place your order and mention the code to our customer service department; 2) Include the code in your fax order; 3) Place your order on the web and enter the promotional code.

Any feedback that you can provide will be greatly appreciated, whether positive or negative. If you have any further questions, please do not hesitate to contact us. We look forward to receiving your Abreview and wish you luck with your research.

The terms and conditions applicable to this offer can be found here: www.abcam.com/collaborationdiscount.

Question 2

Tuesday 14-February-2012

Dear technical team,

Our customer would like to purchase MHC I and II antibodies.

Her sample is mouse DBA II N strain and she is going to test IHC-P.

There are many MHC antibodies so I don't know which one is right product for her.

Please recommend one..

Thank you in advance.

Best regards,

ANSWER:

Thank you for your inquiry.

According to my information the mouse strain DBA/2NCrl has the haplotype H2d. Please encourage your customer strongly to double check this for the strain they mentioned (DBA II N strain).

We do not have any anti MHC class I antibodies that a re tested in IHC-P, but I could offer your customer to take part in the testing discount program for ab64368 that should be suitable for haplotype H2d.

http://www.abcam.com/index.html?datasheet=64368 (or use the following: http://www.abcam.com/index.html?datasheet=64368).

I can also suggest to use an antibody against the beta chain of MHC class I which isnot haplotype dependent: ab75853

http://www.abcam.com/index.html?datasheet=75853 (or use the following: http://www.abcam.com/index.html?datasheet=75853).

This antibody also hs not been tested in IHC-P and we could offer the testing discount to your customer.

For MHC class II we believe that ab15630 might be suitable for it and we could also offer the testing discount.

http://www.abcam.com/index.html?datasheet=15630 (or use the following: http://www.abcam.com/index.html?datasheet=15630).

I am sorry that I could not provide your customer with a definite answer on this occasion and hope the information is nevertheless helpful. Please let me know how your customer would like to proceed and if they would like to take part in the testing discount program.

Question 3

Wednesday 15-September-2010

Why is the recommended dilution for IP (1:40) so different from the recommended dilution for WB (1:10,000- 1:50,000)?

ANSWER:

Thank you for your inquiry.

When performing western blots on immunoprecipitated samples, it is recommended to use the primary antibody 10-25 times more concentrated than that used in western blots on lysates. This is the reason why an antibody used at a dilution of 1:10,000 in western blots on lysates might be used at a dilution of 1:40 in western blots on immunoprecipitated samples.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
