I'm sorry to hear you are having a problem with ab15976. This antibody has only been tested in IHC using formalin fixed, paraffin embedded human tissue. It may very well cross-react with mouse, and it has never been tested on frozen sections or on cytospins, so we do not know if it will work in these applications. Keeping this in mind, however, I would like to suggest the following modifications to your protocol that I believe may reduce this binding if it is non-specific: 1) please try blocking endogenous peroxidases by using 0.3% H2O2 in PBS for 10 minutes before adding the primary antibody, 2) try diluting the antibody to 1:800 and incubating for 30 minutes at room temp, 3) block with 10% normal goat serum, and 4) try a different fixation such as formalin, PFA, methanol or acetone. Ab15976 staining Human normal skin. Staining is localized to cell membrane (and to lesser amount cytoplasmatic compartment).
Left panel: with primary antibody at 1 ug/ml. Right panel: isotype control.
Sections were stained using an automated system DAKO Autostainer Plus , at room temperature. Sections were rehydrated and antigen retrieved with the Dako 3-in-1 AR buffer citrate pH 6.0 in a DAKO PT Link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 minutes. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS), then incubated with primary antibody for 20 minutes, and detected with Dako Envision Flex amplification kit for 30 minutes. Colorimetric detection was completed with diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required.
