Question 2

Friday 28-October-2011

Thanks very much for reviewing this case and offering us the chance to try another b-gal antibody. We would be grateful to receive a vial of AB9361, raised in chick. We’ll just have to find a good secondary antibody. With best regards,

ANSWER:

If Chicken secondary is a problem then I would suggest choosing other beta galactosidase antibodies from the catalogue, with other host e.g. rabbit as ab4761.

I hope this will help.

Question 3

Wednesday 06-December-2006

This anitbody data sheet has an image that shows positive ICC results in mammary cells, but there is no species reactivity listed. Would this antibody be suitable for western blot with mouse mammalian cells?

ANSWER:

Thank you for your enquiry.

This antibody shows no cross-reactivity with other E. coli proteins and can be used for identification and characterization of beta-galactosidase fusion proteins expressed in lambda expression vectors. It was successfully used for immunofluorescence detection of cells transfected with vectors containing the lacZ gene. As long as your cells are transfected with vectors containing the lacZ gene, your Western blot should be successful.

Question 4

Thursday 13-April-2006

I am using a construct that codes for the alpha fragment of beta galactosidase. Will this antibody recognize this?

ANSWER:

Thank you for your enquiry.

I am sorry I can not answer the question; however, I did find the following information in a product specific publication for this clone by Slavickova et al., PubMed ID 1284241. In this publication I have found these data: BG-02 reacted in WB analysis with purified beta-gal, as well as with C-terminal beta-gal fusion proteins. No cross-reactivity with other bacterial proteins was observed. Thus BG-02 enables an identification of recombinant proteins in WB and estimation of their relative molecular weight. The reactivity of BG-02 with N-terminal beta-gal has to be tested. BG-02 also reacted with undenatured fusion proteins in situ during the screening for lambda gt11 recombinants and, therefore, can be used as a positive control for detection of fusion proteins under screening conditions. The BG-02 was successfully used for one-step isolation of fusion proteins from bacterial lysates.

I hope this information helps. Please do not hesitate to contact us if you need anything further.

Question 5

Wednesday 14-September-2005

I have a customer who wants me to confirm that this antibody has successfully been used in immunofluorescence on guinea pig tissue. I know this is mentioned in the datasheet, but not directly and she wants me to confirm this.

ANSWER:

Thank you for your enquiry.

To our knowledge, our beta Galactosidase antibody [BG-02]has yet to be tested by immunohistochemistry or on guinea pig tissue. All tested applications are specified on Abcam product datasheets.

Should you require further assistance, please do not hesitate to contact me.
