Our species is yeast(http://en.wikipedia.org/wiki/Saccharomyces_cerevisiae) and we do reduce, denature the proteins.

We have predicted cross reactivity with this species, but we have not yet tested in Saccharomyces cerevisiae and did not know what results would occur when using this in yeast. Thanks for the response. Actually these blots were fresh and were used only with ab15568(Tubilin from Abcam)   Brief protocol   Gel used                               :              hand cast Tris-HCL 10% Gel condition                     :               100V until ladder reached stacking, then shooted it to 200v until dye past the glass @ Room Temperature Transfer                               :               75V 1hr, cold room(4 degree) Blocking                               :               5% milk Room Temperature 2 hrs Primary antibody             :               ab15568 @ 1:1000, in 1% milk Overnight cold room Secondary antibody        :               Rabbit 1:15000 in 1% milk 45 min Room Temperature   Used ECl kit and developed the films.   Also tried the same protocol with ab15568 @ 1:1500 dilution but both the times I am getting strong 3 bands for the control. I am wondering, why I am seeing GAPDH band so strong when I use Tubilin antibody. All are fresh membranes no stripping at all.   Hope this clarifies and your help is appreciated.

I have just a few more questions regarding this. What species are you testing this product against? Do you run these westerns natively or in a reduced and denatured condition?

Dear Tech support

 

One of our customers is interested in testing ab15568 for WB in two fungi species.

The product's datasheet states the antibody is predicted to react with different Fungi species and the customer wishes to test it for Sclerotinia and Neurospora.

 

Is it possible to test the antibody for above purpose as part of the abreview promotion.

Thank you for your enquiry.

I can confirm that our source has kindly compared the immunogen for ab15568 against the beta tubulin sequence for Sclerotinia (A7EH60) and Neurospora (P05220) and the percentage homology is 92% and 100%, respectively.

These alignments are high, and we would therefore suggest that the antibody is likely to detect in these fungi species. However, as you are aware I am sorry we are not able to guarantee this without further testing. I would be pleased to provide a discount code for the customer. In order to do so, I would appreciate if you could please provide the customers name and also the name of the institution they are working at for our records and so we can trace the Abreview when it is submitted.
