  • Target

    • RelevanceBrevican is the most abundant chondroitin sulfate proteoglycan in the extracellular matrix of the adult brain. It is a member of the lectican family of aggregating extracellular matrix proteoglycans that bear chondroitin sulfate (CS) chains. It is highly expressed in the central nervous system and is thought to stabilize synapses and inhibit neural plasticity. Brevican is secreted from astrocytes and neurons as a 145 kD core protein that bears up to three, covalently-linked, CS chains. It is also is secreted as a 145 kD core protein without CS chains. When cleaved by extracellular glutamyl endopeptidases, the ADAMTSs, a 55 kD N-terminal fragment is formed that contains the unique C-terminal epitope EAMESE.
    • Cellular localizationIsoform 1 is secreted, extracellular space, extracellular matrix; Isoform 2 is found on the membrane; Lipid-anchor, GPI-anchor.

Lane 1 : Brevican transfected 293T cell lysate
      Lane 2 : Non-transfected 293T cell lysate

Lysates/proteins at 25 µg per lane.

      Secondary
Goat Anti-Mouse IgG (H&L)-HRP Conjugate at 1/2500 dilution

Predicted band size : 99 kDa
Observed band size : 170 kDa (why is the actual band size different from the predicted?)
Additional bands at : 125 kDa. We are unsure as to the identity of these extra bands.

    Thank you for contacting Abcam.

You could use Cos7, HeLa, HepG2 cells, or any other cell line to transfect the brevican and use those lysates as a positive control. We only used HEK293T cells as that is what we commonly use, but any other mam...
