As discussed I would suggest trying the following: - Lysing cells in RIPA buffer to ensure sufficient cellular lysis and protein release from the nucleus. - 5% BSA for blocking and primary/secondary antibody incubations. I am interested in potentially using anti-cfos antibody AB16902 in a species of fish. I understand that this antibody has not yet been tested in fish. When I compared the epitope sequence to my fish sequence, it shares 88% identity in this region, with amino acids: kveqmtpeeeekrrirrernkqaaa. Do you think this antibody is likely to cross-react with c-fos protein with this sequence?

Usually we say that anything over 85% homology does have a good chance of cross-reacting, but because we have not tested this species, we can not guarantee that this antibody would work for that species. If you do purchase the antibody and try it in the fish species, please let us know how it goes.
