We are looking for a c-fos antibody that will work in zebrafish whole mount tissue and we need to try a few, so would appreciate if we could receive a sample to test of Anti-c-Fos antibody [2G9C3] (ab16902)

We are obtaining 2 bands of incorrect size in Western blot with ab16902

Bands at 71 and 100 kDa, not at expected size.

Human cell line samples.

Sorry to hear ab16902 is not working well in western blot. As discussed I would suggest trying the following:

- Lysing cells in RIPA buffer to ensure sufficient cellular lysis and protein release from the nucleus. - 5% BSA for blocking and primary/secondary antibody incubations.

I hope it improves the results. If there are still problems, please do let me know and I can replace the product with an alternative.

I am interested in potentially using anti-cfos antibody AB16902 in a species of fish. I understand that this antibody has not yet been tested in fish. When I compared the epitope sequence to my fish sequence, it shares 88% identity in this region, with amino acids: kveqmtpeeeekrrirrernkqaaa. Do you think this antibody is likely to cross-react with c-fos protein with this sequence?

Thank you for your enquiry. Usually we say that anything over 85% homology does have a good chance of cross-reacting, but because we have not tested this species, we can not guarantee that this antibody would work for that species. If you do purchase the antibody and try it in the fish species, please let us know how it goes. I am looking for the blocking peptide for ab16902 antibody. Can I use ab7997 c-fos peptide as a blocking peptide for IHC?

Ab16902 and ab7963 are were raised against different regions of Human c-Fos. Ab16902 was raised against the synthetic peptide: KVEQLSPEEEEKRRIRRERNKMAAA, corresponding to Internal sequence amino acids 128 - 152 of Human c-Fos. Ab7963 was raised against the synthetic peptide: MMFSGFNADYEASS, corresponding to N terminal amino acids 1-14 of Human c-Fos.
