| Product Name | Calpain 1 antibody - Aminoterminal end domain I |
| | See all Calpain 1 antibodies (9)... |
| Product type | Primary antibodies |
| Description | Rabbit polyclonal to Calpain 1 - Aminoterminal end domain I |
| Immunogen | Synthetic peptide based on the amino terminal end of the large subunit of human calpain 1 |
| Reacts with (species key) | Hu, Ms, Rat |
| Specificity | ab28257 recognizes the amino terminal end of the large subunit of Calpain 1. The antibody does not recognize amino-processed Calpain 1, and can be used to discriminate between latent and amino-processed forms. Also, it does not cross react with the other calpain family members (M Calpain, Calpain 94, ncl2, ncl3, etc.). We have a range of domain specific antibodies for this target. For a full list please see all Calpain 1 antibodies |
| Epitope | ab28257 reacts with an epitope located in the first 30 amino acids of the propeptide domain, (Domain I) of Calpain 1, aminoterminal from the activation cleavage site. |

| Application notes (see key) | Recommended dilutions WB: 1/1000, when using colorimetric substrates such as BCIP/NBT - 1/5000, when using chemiluminescent substrates. Predicted molecular weight: 82 kDa. But note that when used against the reduced protein, ab28257 identifies bands at 80 kDa, 68 kDa, and a series of further cleaved forms. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |
| Positive control (see definition) | Human condrosarcoma cell lysate. |
| Cellular localization | Cytoplasmic. Cell membrane. Translocates to the plasma membrane upon Ca(2+) binding. |

| Relevance | Mu Calpain, also known as Calpain I, and m calpain, also known as Calpain II, are intracellular, calcium dependent cysteine proteases. Mu calpain has a micromolar sensitivity (thus the mu) as compared to the millimolar calcium sensitivity of m calpain. The calpains have papain like activity, thus the -pain nomenclature. Both m calpain and mu calpain are ubiquitously expressed, and are countered by the endogenous calpain inhibitor, calpastatin. Other calpain family members (calpain 94, ncl2, ncl3, etc) have more limited tissue distribution, and perhaps different functions. The calpain family members consist of a common small subunit (Calpain 4), and a large variable subunit. It is not clear that all calpains contain a small subunit. Domains in the large subunit include the amino terminal domain I, the proteinase domain II, domain III, and the EF hand domain IV. The calpains, are nonlysosomal, intracellular cysteine proteases. The mammalian calpains include ubiquitous, stomach specific, and muscle specific proteins. The ubiquitous enzymes consist of heterodimers with distinct large, catalytic subunits associated with a common small, regulatory subunit. |
