WB: 1/1000, when using colorimetric substrates such as BCIP/NBT - 1/5000, when using chemiluminescent substrates. Predicted molecular weight: 82 kDa. But note that when used against the reduced protein, ab28258 identifies bands at 80 kDa, 68 kDa, and a series of further cleaved forms. Dilution optimised using Chromogenic detection.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Mu Calpain, also known as Calpain I, and m calpain, also known as Calpain II, are intracellular, calcium dependent cysteine proteases. Mu calpain has a micromolar sensitivity (thus the mu) as compared to the millimolar calcium sensitivity of m calpain. The calpains have papain like activity, thus the -pain nomenclature. Both m calpain and mu calpain are ubiquitously expressed, and are countered by the endogenous calpain inhibitor, calpastatin. Other calpain family members (calpain 94, ncl2, ncl3, etc) have more limited tissue distribution, and perhaps different functions. The calpain family members consist of a common small subunit (Calpain 4), and a large variable subunit. It is not clear that all calpains contain a small subunit. Domains in the large subunit include the amino terminal domain I, the proteinase domain II, domain III, and the EF hand domain IV. The calpains, are nonlysosomal, intracellular cysteine proteases. The mammalian calpains include ubiquitous, stomach specific, and muscle specific proteins. The ubiquitous enzymes consist of heterodimers with distinct large, catalytic subunits associated with a common small, regulatory subunit. |

Immunocytochemistry/ Immunofluorescence - Calpain 1 antibody - Domain I (ab28258) ab28258 staining human HUVEC cells by ICC/IF. Cells were PFA fixed and permeablized in 0.5% Triton X-100 prior to blocking in 1% serum for 16 hours at 4°C. The primary antibody was diluted 1/100 and incubated with the sample for 2 hours at 25°C.
