| Application notes (see key) | Recommended dilutions WB: A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. When used against the reduced protein, Ab38965 identifies bands at 78 Kd, 66 Kd, and a series of further cleaved forms. Many cell types produce RP4Calpain-10; since it is a cellular protein, cell lysate works well for Western blots. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Calpains are ubiquitous, well-conserved family of calcium-dependent, cysteine proteases. The calpain proteins are heterodimers consisting of an invariant small subunit and variable large subunits. The large catalytic subunit has four domains: domain I, the N-terminal regulatory domain that is processed upon calpain activation; domain II, the protease domain; domain III, a linker domain of unknown function; and domain IV, the calmodulin-like calcium-binding domain.
Calpain 10 is an atypical calpain in that it lacks the calmodulin-like calcium-binding domain and instead has a divergent C-terminal domain. It is similar in organization to calpains 5 and 6. It is associated with type 2 or non-insulin-dependent diabetes mellitus (NIDDM) and the gene is located within the NIDDM1 region. Multiple alternative transcript variants encoding different isoforms have been described. |
