| Application notes (see key) | Recommended dilutions WB: 1/1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. Predicted molecular weight: 74.9 kDa. Post-translational modifications such as phosphorylation can produce isoforms with larger apparent MW on SDS PAGE gels. When used against the reduced protein, ab28226 identifies bands at 78 kD, 66 kD, and a series of further cleaved forms. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Calpains are ubiquitous, well-conserved family of calcium-dependent, cysteine proteases. The calpain proteins are heterodimers consisting of an invariant small subunit and variable large subunits. The large catalytic subunit has four domains: domain I, the N-terminal regulatory domain that is processed upon calpain activation; domain II, the protease domain; domain III, a linker domain of unknown function; and domain IV, the calmodulin-like calcium-binding domain.
Calpain 10 is an atypical calpain in that it lacks the calmodulin-like calcium-binding domain and instead has a divergent C-terminal domain. It is similar in organization to calpains 5 and 6. It is associated with type 2 or non-insulin-dependent diabetes mellitus (NIDDM) and the gene is located within the NIDDM1 region. Multiple alternative transcript variants encoding different isoforms have been described. |
