| Application notes (see key) | Recommended dilutions WB: A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. When used against the reduced protein this antibody identifies bands at 75 kD, 58kD, and a series of further cleaved active forms. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Calpains are ubiquitous, well-conserved family of calcium-dependent, cysteine proteases. The calpain proteins are heterodimers consisting of an invariant small subunit and variable large subunits. The large subunit possesses a cysteine protease domain, and both subunits possess calcium- binding domains. Calpains have been implicated in neurodegenerative processes, as their activation can be triggered by calcium influx and oxidative stress. Calpain 6 is highly expressed in the placenta. Its C-terminal region lacks any homology to the Calmodulin-like domain of other Calpains. It also lacks critical active site residues and thus is suggested to be proteolytically inactive. |
