| Application notes (see key) | Recommended dilutions WB: 1/1000 when using colorimetric substrates such as BCIP/NBT, and 1/5000 for chemiluminescent substrates. Detects a band of approximately 30kDa and 28kDa (predicted molecular weight: 30 kDa). Calpain S1 is quickly degraded, and samples must be lysed in the presence of protease inhibiters to maintain latency. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Calpains are a ubiquitous, well-conserved family of calcium-dependent, cysteine proteases. Calpain families have been implicated in neurodegenerative processes, as their activation can be triggered by calcium influx and oxidative stress. Calpain I and II are heterodimeric with distinct large subunits associated with common small subunits, all of which are encoded by different genes. Calpain S1 is a small subunit common to both Calpain I and II and is associated with myotonic dystrophy. |
