Mental retardation is characterized by significantly below average general intellectual functioning associated with impairments in adaptative behavior and manifested during the developmental period. Patients with mental retardation X-linked CASK-related can manifest a severe phenotype consisting of severe intellectual deficit, congenital or postnatal microcephaly, disproportionate brainstem and cerebellar hypoplasia (MICPCH Syndrome). A milder phenotype consists of mental retardation alone or associated with nystagmus.
    Defects in CASK are the cause of FG syndrome type 4 (FGS4) [MIM:300422]. FG syndrome (FGS) is an X-linked disorder characterized by mental retardation, relative macrocephaly, hypotonia and constipation.

Anti-CASK antibody [S56A-50] images

  • Predicted band size : 103 kDa
    Additional bands at : 110 kDa. We are unsure as to the identity of these extra bands.

    Lane 1: Wild-type HAP1 cell lysate (20 µg)
    Lane 2: CASK knockout HAP1 cell lysate (20 µg)
    Lane 3: Rat brain tissue lysate (20 µg)
    Lane 4: SH-SY5Y cell lysate (20 µg)
    Lanes 1 - 4: Merged signal (red and green). Green - ab99039 observed at 110 kDa. Red - loading control, ab18251, observed at 50 kDa.

    ab99039 was shown to specifically react with CASK when CASK knockout samples were used. Wild-type and CASK knockout samples were subjected to SDS-PAGE. ab99039 and ab18251 (loading control to alpha Tubulin) diluted to 1/500 and 1/10000 respective were incubated overnight at 4°C. Blots were developed with goat anti-rabbit IgG (H + L) and goat anti-mouse IgG (H + L) secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

  • Overlay histogram showing SH-SY5Y cells stained with ab99039 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab99039, 1μg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 1μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.
