The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 0.5 - 1 µg/ml. Predicted molecular weight: 118 kDa.
Use a concentration of 5 µg/ml.
FunctionInactive tyrosine kinase involved in Wnt signaling pathway. Component of both the non-canonical (also known as the Wnt/planar cell polarity signaling) and the canonical Wnt signaling pathway. Functions in cell adhesion, cell migration, cell polarity, proliferation, actin cytoskeleton reorganization and apoptosis. Has a role in embryogenesis, epithelial tissue organization and angiogenesis.
Tissue specificityHighly expressed in lung, liver, pancreas, kidney, placenta and melanocytes. Weakly expressed in thyroid gland, ovary, brain, heart and skeletal muscle. Also expressed in erythroleukemia cells. But not expressed in colon.
Sequence similaritiesBelongs to the protein kinase superfamily. Tyr protein kinase family. Insulin receptor subfamily. Contains 7 Ig-like C2-type (immunoglobulin-like) domains. Contains 1 protein kinase domain.
DomainThe protein kinase domain is predicted to be catalytically inactive.
Post-translational modificationsMMP14 cleaves PTK7 between Pro-621 and Leu-622 generating an N-terminal soluble (70 kDa) fragment and a membrane C-terminal (50 kDa) fragment. Proteolysis by MMP14 regulates PTK7 function in non-canonical Wnt signaling pathway.
Cellular localizationMembrane. Cell junction. Colocalizes with MMP14 at cell junctions. Also localizes at the leading edge of migrating cells.

ab62074 staining CCK4 in E9.5 mouse embryo tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 in PBS and blocked with 5% serum for 1 hour at 25°C. Samples were incubated with primary antibody (1/500 in 5% donkey serum) for 12 hours at 4°C. An Alexa Fluor® 488-conjugated donkey anti-rabbit IgG polyclonal (1/1000) was used as the secondary antibody.
Image shows sagittal section of E9.5 mouse embryo. Neuroectoderm and underlying mesoderm labelled with ab62074 in green. DAPI stained nuclei in blue.
