The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
Flow Cyt Use at an assay dependent concentration. ab171870-Rabbit polyclonal IgG, is suitable for use as an isotype control with this antibody.
IHC-P 1/100. For antigen retrieval boil tissue section in 10mM Citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.
WB Use a concentration of 1 µg/ml. Detects a band of approximately 100 kDa (predicted molecular weight: 86 kDa).

Target

  • FunctionThermolysin-like specificity, but is almost confined on acting on polypeptides of up to 30 amino acids. Biologically important in the destruction of opioid peptides such as Met- and Leu-enkephalins by cleavage of a Gly-Phe bond. Able to cleave angiotensin-1, angiotensin-2 and angiotensin 1-9. Involved in the degradation of atrial natriuretic factor (ANF). Displays UV-inducible elastase activity toward skin preelastic and elastic fibers.
  • Sequence similaritiesBelongs to the peptidase M13 family.
  • Cellular localizationCell membrane.

  • Anti-CD10 antibody (ab82073) at 1 µg/ml + Human kidney tissue lysate - total protein (ab30203) at 10 µg

    Secondary
Goat Anti-Rabbit IgG H&L (HRP) preadsorbed (ab97080) at 1/5000 dilution
    Developed using the ECL technique

    Performed under reducing conditions.

Predicted band size : 86 kDa
Observed band size : 100 kDa (why is the actual band size different from the predicted?)

    Exposure time : 1 minute
CD10 contains a number of potential glycosylation sites (SwissProt), which may explain its migration at a higher molecular weight than predicted.
